I second the Out of the Kitchen recommendation. Their cupcakes are the most consistently moist; Magnolia's are dryer than your average lump of sand.

Buttercup is a spinoff/splinter of Magnolia. It is MUCH better in my opinion - the cupcakes tend to be fresher and there is a wider selection of flavors. I do feel Magnolia is a bit overrated. Too sweet and too often slightly stale :P Cupcake Cafe is my current favorite...

Still think 'Out Of The Kitchen' on Hudson Street in West Village is a much overlooked but fantastic place to buy your cakes.

I'm so glad Passover is over so I can eat lots of cupcakes! Apparently there is a new bakery on Rivington called Sweet something with great cupcakes...I don't know the name cause I haven't been there yet. Magnolia will always be my favorite but they need to open another branch of it so it's more easily accessible. Thanks for the consistant cupcake posting. Of course, that makes me crave them all day, but I forgive you for that.
